WHAT IS THE ORNITHINE CYCLE?

The ornithine cycle (or otherwise known as the urea cycle) is a cycle of ammonia degradation. Otherwise known as: the urea cycle or the urea Krebs cycle.

The urea cycle is the primary metabolic pathway during which ammonia is degraded and urea is synthesized. Ammonia undergoes transformation to individual amino acids to eventually become urea, which will be excreted from the body with urine, and in small amounts with sweat. Individual ammonia metabolism requires, among other things, two amino acids: ornithine and aspartate, as well as cellular energy ATP.

HOW DOES THE ORNITHINE CYCLE PROCEED?

The urea cycle is a sequence of consecutive enzymatic reactions of three amino acids: ornithine, citrulline, and arginine.

The metabolic cycle of the above amino acids produces urea from ammonia, carbon dioxide, and aspartate.

In fact, from the decomposition of the amino acid ornithine, after transferring the amino (-NH2) residue, four molecules are formed by means of transamination reactions: a-ketoglutarate, glutamate, oxaloacetate or pyruvate.

These amino acids are secreted into the blood and then enter the liver, where the transamination reaction involves alanine together with aspartate, and their amino groups are transferred to α-ketoglutarate with the formation of glutamate or to glutamate with the formation of glutamine. Glutamate then undergoes a chemical reaction that strips it of its amino group and the resulting ammonium ion is immediately converted into carbamoylphosphate. The latter is incorporated into the ornithine cycle.

The ammonium ion is a highly toxic substance, so it must be degraded into a water-soluble and less toxic substance, urea. It gives the human body a chance to get rid of all the by-products and unnecessary products of metabolism.

WHERE DOES THE ORNITHINE CYCLE OCCUR?

The entire ornithine cycle takes place in the mitochondria and liver cells of hepatocytes. It is directly connected to the Krebs cycle proper through the electron transport chain, adenosine triphosphate (ATP) as the cell's energy carrier, and fumaric and aspartic acids. The main product of metabolism is urea.
